Abstract

Proposed review-derived manuscript investigating tumor immune-escape mechanisms through single-cell CRISPR perturbation mapping, spatial immunology, and predictive response modeling for immune checkpoint blockade. The study synthesizes evidence from CRISPR screens, single-cell profiling, and computational frameworks to identify multicellular immune resistance networks, including T-cell exhaustion, immune exclusion, antigen presentation failure, and myeloid suppression.
